Research Background:
Tirzepatide is a synthetic 39-amino-acid peptide engineered as a dual agonist of the glucose-dependent insulinotropic polypeptide (GIP) receptor and the glucagon-like peptide-1 (GLP-1) receptor. Its sequence is based on the native GIP backbone and includes two 2-aminoisobutyric acid (Aib) substitutions for DPP-4 resistance, along with a C20 fatty diacid moiety attached through a linker to promote albumin binding and extended half-life. Preclinical and in vitro research has investigated its receptor pharmacology, including binding affinity at GIP and GLP-1 receptors, downstream cAMP signaling, and biased agonism at the GLP-1 receptor. Laboratory studies have also examined incretin receptor co-agonism in cellular and animal models, including interactions with pancreatic islet signaling pathways.
